Ph. Eur. 12.3 strengthens the role of TOC testing in pharmaceutical water quality control. This article explores the latest changes and how Online TOC Analyzer technology can help manufacturers achieve real-time monitoring, contamination control, and reliable data management for WFI and purified water systems.
On July 1, 2026, Issue 12.3 of the 12th Edition of the European Pharmacopoeia (Ph. Eur.) officially came into effect, introducing revisions to three texts on water for pharmaceutical use: Water for Injections (WFI, 0169), Purified Water (0008), and general chapter Total Organic Carbon in Water for Pharmaceutical Use (2.2.44).
The headline change is narrower than it is often reported: in the Sterilised Water for Injections (SWFI) section of monograph 0169, the test for oxidisable substances has been replaced by the more sensitive and non-selective Total Organic Carbon (TOC) test. Bulk WFI and bulk Purified Water were already controlled by TOC; for them, the 12.3 changes are clarifications rather than new requirements.
The revision goes further than a single test substitution. General chapter 2.2.44 has been expanded, and four points matter in practice:
One point is easily missed but decisive for water system design: chapter 2.2.44 allows TOC to be determined on-line as well as off-line. A qualified online analyzer can therefore be used to demonstrate pharmacopoeia compliance, not only to support process monitoring.
Compared with traditional testing methods, TOC analysis provides a more comprehensive evaluation of organic impurities with higher sensitivity and better suitability for continuous quality monitoring. TOC is now recognized across the major pharmacopoeias, including USP, JP, IP and Ph. Eur., and the Ph. Eur. revision is an explicit step towards a global quality standard for SWFI, the most widely used excipient in the industry.
For sterile pharmaceutical manufacturers, establishing a stable and reliable TOC monitoring system is essential for strengthening water quality control and contamination risk management.
Traditional pharmaceutical water TOC testing is usually performed through manual sampling followed by laboratory analysis. Although this approach can meet routine testing requirements, it may present limitations for continuously operating pharmaceutical water systems:
For critical water systems such as WFI distribution loops, continuous, near-real-time monitoring provides continuous visibility into water quality trends. An Online TOC Analyzer enables manufacturers to monitor organic contamination changes continuously, helping identify potential risks earlier and improve overall process control.
To meet the increasing demand for reliable pharmaceutical water monitoring, Tailin developed the DI1500-OL Online Total Organic Carbon (TOC) Analyzer.
Designed for critical applications including WFI and purified water systems, the analyzer combines high-sensitivity detection, flexible system integration, low-maintenance operation, and full data traceability, providing a precise, efficient, and GMP-ready pharmaceutical water monitoring solution.
Pharmaceutical water systems require TOC analyzers with high sensitivity, excellent stability, and long-term reliability.
The Tailin DI1500-OL Online TOC Analyzer offers:
The analyzer adopts a maintenance-friendly modular design. Key components are easy to maintain, and consumables can be replaced independently. Without frequent reagent replacement, the DI1500-OL significantly reduces operating and maintenance costs while improving system efficiency.
The DI1500-OL supports multiple control configurations, including PC-based control; Four-channel transmitter control; Industrial computer (IPC) control.

The analyzer is equipped with multiple analog signal (4–20 mA) and digital signal output channels as standard, enabling integration with water distribution systems. This allows contamination trends to be identified quickly and enables the water system to trigger actions such as flow shut-off, diversion, and discharge.
Modern pharmaceutical quality control requires not only accurate analytical results but also complete data traceability.
The DI1500-OL Online TOC Analyzer supports:
The system is designed to support compliance with FDA 21 CFR Part 11, EU GMP Annex 11, and ALCOA+ data integrity requirements, providing reliable records for quality audits and regulatory inspections. In addition, Tailin provides comprehensive IQ/OQ/PQ validation services, together with calibration and system suitability testing in line with Ph. Eur. 2.2.44 and USP <643> , to help pharmaceutical manufacturers establish compliant TOC monitoring systems aligned with sterile drug manufacturing requirements.
With Ph. Eur. 12.3 now in force and TOC established as the reference method for organic impurities in pharmaceutical water, online TOC analyzer technology is becoming an important tool for continuous water quality monitoring and for the contamination control strategies required under EU GMP Annex 1.
With accurate detection, reliable operation, and advanced data management capabilities, the Tailin DI1500-OL Online TOC Analyzer provides pharmaceutical companies with an effective solution for WFI and purified water monitoring, supporting safer and more compliant sterile pharmaceutical production.
