Global Pharmacopoeias Drive the Advancement of TOC Testing Standards

On July 1, 2026, Issue 12.3 of the 12th Edition of the European Pharmacopoeia (Ph. Eur.) officially came into effect, introducing revisions to three texts on water for pharmaceutical use: Water for Injections (WFI, 0169), Purified Water (0008), and general chapter Total Organic Carbon in Water for Pharmaceutical Use (2.2.44).

The headline change is narrower than it is often reported: in the Sterilised Water for Injections (SWFI) section of monograph 0169, the test for oxidisable substances has been replaced by the more sensitive and non-selective Total Organic Carbon (TOC) test. Bulk WFI and bulk Purified Water were already controlled by TOC; for them, the 12.3 changes are clarifications rather than new requirements.

The revision goes further than a single test substitution. General chapter 2.2.44 has been expanded, and four points matter in practice:

  • Method B has been introduced for SWFI, with three TOC acceptance limits applied according to container size;
  • Method A is now explicitly referenced for bulk WFI and bulk Purified Water;
  • The TOC limit is now written as 0.50 mg/L rather than 0.5 mg/L, aligning it with the concentration of the standards used in the chapter;
  • Sucrose R and 1,4-benzoquinone R have been replaced by chemical reference substances (CRSs) to streamline the system suitability test, and the conductivity test has been clarified so that the permitted tolerance refers to the expected conductivity of the reference solution rather than the measured value.

One point is easily missed but decisive for water system design: chapter 2.2.44 allows TOC to be determined on-line as well as off-line. A qualified online analyzer can therefore be used to demonstrate pharmacopoeia compliance, not only to support process monitoring.

Compared with traditional testing methods, TOC analysis provides a more comprehensive evaluation of organic impurities with higher sensitivity and better suitability for continuous quality monitoring. TOC is now recognized across the major pharmacopoeias, including USP, JP, IP and Ph. Eur., and the Ph. Eur. revision is an explicit step towards a global quality standard for SWFI, the most widely used excipient in the industry.
